| I. Background | |
| CD40 是腫瘤壞死因子受體超家族（TNFRSF）成員，作為免疫調(diào)節(jié)的關(guān)鍵分子，廣泛 參與 B 細(xì)胞活化、抗原呈遞細(xì)胞（APC）成熟及 T 細(xì)胞免疫應(yīng)答。其配體 CD40L（CD154） 主要表達(dá)于活化的 T 細(xì)胞，通過 CD40-CD40L 互作橋接先天性與適應(yīng)性免疫，是腫瘤免疫 治療、自身免疫病及感染性疾病的熱門靶點(diǎn)。 CD40 信號(hào)轉(zhuǎn)導(dǎo)主要利用 TNF 受體相關(guān)因子的銜接蛋白，導(dǎo)致經(jīng)典和非經(jīng)典 NFκB 通路、MAPK、PI3K 和 PLCγ的激活。它們的激活導(dǎo)致特征性下游效應(yīng)，包括轉(zhuǎn)錄激活、細(xì) 胞骨架重排和細(xì)胞存活。 |

CD40 NFκB-Luc HEK293 報(bào)告基因藥靶模型很好的模擬了體內(nèi) CD40 的信號(hào)轉(zhuǎn)導(dǎo)過程， 原理見下圖所示。
